Semaglutide is a long-acting analogue of glucagon-like peptide-1, commonly known as GLP-1. It shares approximately 94% sequence similarity with human GLP-1 and has been modified to resist enzymatic breakdown and remain active in circulation for longer.
Studied Uses
Semaglutide has been clinically studied in areas including:
- Type 2 diabetes and blood-glucose regulation
- Obesity and long-term weight management
- Appetite and caloric-intake regulation
- Cardiovascular-risk reduction
- Kidney outcomes in people with type 2 diabetes and chronic kidney disease
- Noncirrhotic metabolic dysfunction-associated steatohepatitis, or MASH, with moderate-to-advanced liver fibrosis
Authorized semaglutide formulations have specific indications depending on the product and formulation. Clinical research has examined its effects on glycaemic control, body weight, cardiovascular outcomes, kidney-disease progression and liver-related metabolic disease.
How It Works
Semaglutide selectively binds to and activates GLP-1 receptors. When blood glucose is elevated, this signalling increases insulin secretion and reduces glucagon secretion, helping regulate blood-glucose levels.
GLP-1 receptors are also present in areas of the brain involved in appetite regulation. Their activation can reduce hunger and caloric intake. Semaglutide additionally delays gastric emptying, which affects how quickly food and glucose leave the stomach.
