Melanotan-1, also known as afamelanotide, is a synthetic 13-amino-acid analogue of alpha-melanocyte-stimulating hormone, or α-MSH. It has been investigated for its effects on melanin production, skin pigmentation and biological responses to light exposure.
Studied Uses
Melanotan-1 and afamelanotide have been studied in research involving:
- Melanin production and skin pigmentation
- Photoprotection and responses to ultraviolet exposure
- Painful photosensitivity associated with erythropoietic protoporphyria
- Repigmentation in vitiligo when combined with narrowband UV-B phototherapy
- Melanocortin-1 receptor signalling in human skin
Early clinical studies found that Melanotan-1 increased skin pigmentation in human participants. The approved afamelanotide implant has also been clinically studied and authorized to increase pain-free light exposure in adults with erythropoietic protoporphyria.
Research has additionally examined afamelanotide alongside narrowband UV-B phototherapy for repigmentation in people with nonsegmental vitiligo.
How It Works
Melanotan-1 acts primarily as an agonist of the melanocortin-1 receptor, or MC1R, found on melanocytes. Activation of this receptor stimulates the production of eumelanin, the brown-black pigment involved in skin pigmentation and photoprotection.
The resulting increase in eumelanin can darken skin pigmentation and reduce the penetration of certain wavelengths of light. Research has also examined MC1R signalling in relation to antioxidant activity, DNA-repair responses and inflammatory regulation.
